摘要:
目的:对葡萄球菌性烫伤样皮肤综合征(SSSS)快速诊断试剂盒的研发进行探索。方法:采用表皮剥脱毒素A抗体(Anti-ETA)、表皮剥脱毒素A(ETA),优化反应条件后将Anti-ETA包被ELISA板,初步建立SSSS表皮剥脱毒素A的ELISA检测方法。结果:在建立的ELISA方法中,Anti-ETA的最佳包被浓度为10 μg·mL-1,酶标二抗最佳工作浓度为1:100。对7份SSSS患者血清、7份内毒素阳性患者血清及7份正常人血清进行吸光度值(OD 450 nm)的检测,结果显示SSSS患者血清组和正常人血清组的OD值无差别(P>0.05),即单纯包被Anti-ETA尚不能作为SSSS的有效检测。结论:本试验初步建立以Anti-ETA作为包被抗体检测SSSS表皮剥脱毒素A的ELISA方法,但单纯包被Anti-ETA检测灵敏度低,研制SSSS的快速诊断试剂盒需进一步结合Anti-ETB作为共同包被抗体。
Abstract:
Objective: To explore the development of a rapid diagnostic kit for staphylococcal scalded skin syndrome(SSSS).Methods: Epidermal exfoliation toxin A antibody(Anti-ETA)and Epidermal exfoliation toxin A(ETA)were used.After optimizing the reaction conditions, Anti-ETA was coated on ELISA plate to established an ELISA assay for detecting ETA.Results: In the established ELISA method, the optimum coating concentration of Anti-ETA was 10 μg·mL-1, and the optimum working concentration of ELISA was 1:100.Hypothesis tests were carried on the OD values of 7 SSSS patient sera, 7 endotoxin-positive patient sera, and 7 normal human sera.The results showed that there was no difference in OD value between SSSS patients and normal persons(P>0.05), that is to say, anti-ETA alone could not be used as an effective detection of SSSS.Conclusions: An ELISA method for detecting SSSS epidermal exfoliation toxin A using Anti-ETA as coated antibody was preliminarily established in this experiment.However, the sensitivity of detecting SSSS epidermal exfoliation toxin A by anti-ETA alone is low.The development of a rapid diagnostic kit for SSSS requires further combination with Anti-ETB as a common coated antibody.
